Principle of the MTT Assay
Figure 1. The MTT assay is a colorimetric method used to assess cell viability and cytotoxicity based on cellular metabolic activity. The yellow compound MTT is reduced by mitochondrial succinate dehydrogenase (SDH) in metabolically active cells to form insoluble purple formazan crystals. These crystals are subsequently dissolved in an organic solvent such as DMSO, yielding a purple solution. The absorbance measured at 550–570 nm is directly proportional to the number of viable cells, thereby reflecting cell viability and proliferation.
Storage
MTT Reagent should be stored at 2-8 °C and shielded from light, and Dimethyl Sulfoxide (DMSO) should be stored at room temperature.
